EFFECTS OF GRAPE SEED PROANTHOCYANIDIN EXTRACT ON OXIDATIVE STRESS AND ANTIOXIDANT ENZYMES IN KIDNEY OF RATS WITH DIABETIC NEPHROPATHY

Abstract. Diabetic nephropathy (DN) is one of the most important micro vascular complications of diabetes. Our aim was to evaluate effects of grape seed proanthocyanidin extract (GSPE) on oxidative stress and activities of antioxidant enzymes in diabetic nephropathy rats. A total of 40 rats were used and were divided into four groups of 10 rats each: control, diabetic, diabetic nephropathy, diabetic nephropathy who’s received 500 mg/kg body weight/ day GSPE for 6 week. The first, 24-hour urinary albumin excretion (UAE) was studied two weeks after induction of diabetes. Then the UAE was studied each week until the end of the experimental period in all of groups. Lipid peroxidation content and activities of Catalase (CAT), Superoxide Dismutase (SOD) and Glutathione Peroxidase (GPx) were measured in kidney homogenate supernatants. The UAE in diabetic nephropathy group were significantly higher than in control and diabetic groups (P < 0.05). An increase of lipid peroxidation content and decrease of CAT, SOD, and GPx activities in kidney of diabetic nephropathy and diabetic rats were observed (P < 0.05). The GSPE administration significantly decreased kidney weight, UAE, lipid peroxidation and augmented the activities of antioxidant enzymes studied in kidney of diabetic nephropathy rats. The GSPE could ameliorate nephropathy in diabetic rats through reduction of oxidative stress and increase of renal antioxidant enzymes activity.
